Abstract : Lead (Pb) is highly poisonous and is largely utilized by industries. It produces nephrotoxicity with notable changes in renal architecture and function. Alhagi Maurorum (AM) belongs to the Fabaceae family and is native to the Middle East, North Africa, Southeast Europe, and Asia. Phenolic compounds namely, orientin, rutin, and quercetin were isolated from AM and these compounds exhibit various pharmacological properties. In this study, we evaluated the potential protective activities of AM in Pb intoxicated rats. Animals were classified into five groups: control, AM (200 mg kg-1, orally), Pb (15 mg kg-1, orally), Pb+AM (100 mg kg-1) and Pb + AM (200 mg kg-1) respectively were treated for 28 days. Pb administration markedly increased serum cystatin C, urea, creatinine levels, and urinary NAG. Pb also caused renal histological alterations. In addition, Pb-treated rats exhibited marked accumulation of MDA, GSH depletion, and CAT and SOD exhaustion. However, Pb intoxication increases Nrf2 and HO-1 expression and markedly induced inflammatory marker (IL-6, TNF-α, COX-2, and NFκB) levels in renal tissue. Pb significantly upregulated Bax and markedly decrease Bcl-2 protein expression. Pb exposure was reported to inhibit the expression of Nrf2 and HO-1 in the kidneys. Thus, inhibition of Nrf2 and HO-1 following PbAc intoxication may also explain the inhibition of antioxidant molecules in this study. the up-regulated Nrf2 and HO-1 expression following AM to be the reason behind the enhanced levels of the antioxidant molecules in the renal tissue. AM treatment exhibits marked kidney protection and ameliorates pathological changes. Conclusively, AM offers protection in contrast to Pb toxicity via antioxidant, anti-inflammatory, and anti-apoptotic mechanisms.
