Abstract
:
Rhazya stricta is medicinal plant used in the treatment of many diseases such as diabetes, blood pressure, sore throat and other diseases. Many studies on Rhazya stricta knowledge has its effect on different functions in organisms such as its impact on biochemical variables, and nervous system. In addition, several studies evaluate genetic toxicity of aqueous extract of Rhazya stricta leaves on several organism such as micro-organisms (Saccharomyces cerevisiae, Aspergillus terreus), rat, mice and also on the human lymphocytes. The aims of this study to evaluate the effects of Rhazya stricta extract on the rats genome at the cytogenetic, molecular and biochemical levels to assess the genotoxicity of this extract. In the present study, aqueous extract of Rhazya stricta produced significant (P< 0.01 -0.05) effects on glucose levels as well as kidney function profile concentrations including uric acid and Creatinine. Rhazya stricta extract did not show any significant effects on the activity of liver enzymes and lipid profile. At cytogenetic level, the result of micronucleus test showed that there is a formation of micronuclei in reticulocytes blood cells for treated rat during the weeks of the experiment. At molecular level, RAPD results showed that polymorphic band were obtained in treated groups comparing to control group with all RAPD primers which were used. All these results strongly point out the mutagenic and Clastogenic effects of the leaves aqueous extract of Rhazya stricta.
